Protocol 44156: Standard PCR Assay - Dock7<m>
Version 6.0

Notes

The way this breeding is done – we get three different phenotypes - db homs are fat and black, Dock 7 or misty homs are lean with a misty coat color & hets are black and lean. So, based on eliminating the two hom classes by phenotype, the only animals that will be genotyped/breeder confirmed are double hets, as we see in all these reqs- From Marge

The genotyping protocol(s) presented here have been optimized for reagents and conditions used by The Jackson Laboratory (JAX). To genotype animals, JAX recommends researchers validate the assay independently upon receipt of animals into their facility. Reaction cycling temperature and times may require additional optimization based on the specific genotyping reagents used.

Expected Results

Mutant = 200 bp
Heterozygote = 200 bp and 481 bp
Wild Type = 481 bp

JAX uses a very high speed Taq (~1000 bp/sec), use cycling times recommended for your reagents.
JAX uses a 'touchdown' cycling protocol and therefore has not calculated the optimal annealing temperature for each set of primers.
