Conditions of use

JAX® Mice, Product & Services conditions of use

"MICE" means mouse strains, their progeny derived by inbreeding or crossbreeding, unmodified derivatives from mouse strains or their progeny supplied by The Jackson Laboratory ("JACKSON"). "PRODUCT(S)" means biological materials supplied by JACKSON, and their derivatives. "SERVICES" means projects conducted by JACKSON for other parties that may include but are not limited to the use of MICE or PRODUCTS. "RECIPIENT" means each recipient of MICE, PRODUCTS, or SERVICES provided by JACKSON including each institution, its employees and other researchers under its control. MICE or PRODUCTS shall not be: (i) used for any purpose other than the internal research, (ii) sold or otherwise provided to any third party for any use, or (iii) provided to any agent or other third party to provide breeding or other services. Acceptance of MICE, PRODUCTS or SERVICES from JACKSON shall be deemed as agreement by RECIPIENT to these conditions, and departure from these conditions requires JACKSON’s prior written authorization.

MICE, PRODUCTS AND SERVICES ARE PROVIDED "AS IS." Credit for products or services

In case of dissatisfaction for a valid reason and claimed in writing by a purchaser within ninety (90) days of receipt of PRODUCTS or SERVICES, JACKSON will, at its option, provide credit or replacement for the PRODUCT received or the SERVICES provided; JACKSON makes no other representations, and this shall be the exclusive remedy of the purchaser. How is germline competency determined?

What data do you have on the germline competency of the B6-693 mES cells?

B6-693 ES cells from have proven germ line competent in six experiments at passages 13, 15, 19, 31 and 32. Four of the six experiments were following ES cell targeting. Chimeric mice were obtained by unilateral transfer of 10-12 microinjected blastocysts to the uteri of ~10 pseudopregnant female mice for each experiment. In six experiments with B6 mES cells, the number of coat-color chimeras divided by the number of mice born were 10/66, 4/13, 6/15, 5/20, 21/59, 10/39. Why has the passage number for the B6-693 mES cells changed over time?

We first distributed the B6-693 mES cells at P16. The batch was expanded from a starting population of ES cells at P12. We generated a limited number of vials at P13 and P14 for seed stock and then expanded to P16 to obtain sufficient quantities of the sellable stock.  We are now distributing  B6-693 mES cells at P11 that were generated with cells from a starting population at P9. This P11 line has been karyotyped male with high percentage of normal spreads.

Why is the earlier passage number advantageous?

mES cells at an earlier passage number typically have higher frequencies of germline transmission and a lower likelihood of chromosomal gains, losses, or translocations (i.e. more likely to have a normal karyotype). In general, the more passages an ES cell line is subjected to, the greater the chance of changes in karyotype that could negatively impact their ability to create germline chimeras.

Why is MEK inhibitor added to the cell culture media?

The MEK inhibitor was included to ensure that cells remained undifferentiated during expansion.

JAX® ES cell development program

Through our ES cell development program, we are generating new tools for both gene targeting and in vitro disease modeling. We have developed methods to produce ES cell lines from recalcitrant inbred strains and mouse models.

To date we have derived ES cell lines from DBA/2J, NSG - NOD-scid IL2 receptor gamma (NOD.Cg-Prkdcscid Il2rgtm1Wjl/SzJ), NRG — NOD Rag IL2 receptor gamma (NOD.Cg-Rag1tm1MomIl2rgtm1Wjl/SzJ) (an alternative to NSG), NOD/ShiLtJ, and C57BL/6J mice.

Most recently, we created ES cells from three of the most widely used Alzheimer’s disease models.  We also introduced a transgene that allows for GFP expression upon neuronal differentiation, which will ease assessment of neuronal development in vitro.

Our derivation methods utilize 2i media, and the most robust emergent ES cell lines are selected based on NANOG expression and euploidy. Where appropriate, our ES cell lines are also germline tested. Ultimately, our fully characterized ES cell lines are made available to the scientific community through Mice & Services®JAX or through the Mutant Mouse Research and Resource Centers (MMRRC).

JAX® Deltagen knockout ES cell lines

The ES cell lines created and characterized by Deltagen Inc. are available from The Jackson Laboratory*. Companion mouse strains created using Deltagen’s ES cell lines are also available via The Jackson Laboratory Repository.

*These products became available after 2005, when the NIH announced an agreement allowing the distribution of mutant mice and frozen embryos from two private collections of knockout (KO) mice.

Research institutions require an Academic Use License, and companies require a license prior to shipping.  

This mouse strain (including any and all progeny, "MICE") is distributed by The Jackson Laboratory with the following terms and conditions.

MATERIALS ARE AVAILABLE TO NONPROFIT RESEARCH INSTITUTIONS, UNIVERSITIES AND HOSPITALS THAT HAVE AN ACADEMIC LICENSE AGREEMENT WITH DELTAGEN INC.  COMPANIES REQUIRE A SEPARATE COMMERCIAL USE LICENSE WITH DELTAGEN.  AN ACADEMIC LICENSE AGREEMENT WILL BE FORWARDED FOR APPROVAL THE FIRST TIME A MATERIAL IS ORDERED.

MATERIALS may be sent by the NIH-designated repositories to any academic or nonprofit organization worldwide that have signed the Academic License with Deltagen, but may not be sent to commercial organizations without a license.
