Description

Tcf4^CreERT2 knock-in/knock-out mice express an inducible Cre/ERT2 fusion protein in connective tissue fibroblasts.

Detailed description

Tcf4^CreERT2 knock-in/knock-out mice have a creERT2 fusion protein replacing exon 1 of the transcription factor 7 like 2, T cell specific, HMG box gene (Tcf7l2). Tcf7l2 is a transcription factor that plays a role in the Wnt signaling pathway and is involved in the maintenance of the epithelial stem-cell compartment of the small intestine. Mutations in this gene have been associated with colon carcinogenesis. Expression is also evident in connective tissue fibroblasts. Heterozygous mice are viable and fertile, while homozygous mice are not viable. When Tcf4^CreERT2 are bred with mice containing loxP-flanked sequence, tamoxifen-inducible Cre-mediated recombination will result in deletion of the floxed sequences in the Cre-expressing cells of the offspring.

Development

A targeting construct was designed to replace the exon 1 of the transcription factor 7 like 2, T cell specific, HMG box gene (Tcf7l2) with a creERT2 fusion gene (a Cre recombinase fused to a human estrogen receptor ligand binding domain) followed by a polyadenylation signal). A frt-flanked neomycin resistance (neo) cassette was inserted downstream of the creERT2 fusion gene. The construct was electroporated into (129X1/SvJ x 129S1/Sv)F1-Kitl⁺-derived R1-45 embryonic stem (ES) cells and correctly targeted ES cells were injected into blastocysts. Chimeric mice were bred to ACTB:FLPe mice (Stock No. 005703) to delete the neo cassette. Resulting Tcf4^CreERT2 mice were subsequently bred to C57BL/6J mice (Stock No. 000664) for six generations. Frozen sperm was shipped to The Jackson Laboratory. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes.
