Detailed description

Tcf4^Cre knock-in/knock-out mice have an EGFP/cre fusion protein replacing exon 1 of the transcription factor 7 like 2, T cell specific, HMG box (Tcf7l2) gene. The donating investigator reports that homozygous mice are not viable. Tcf7l2 is a transcription factor that plays a role in the Wnt signaling pathway and is involved in the maintenance of the epithelial stem-cell compartment of the small intestine. Mutations in this gene have been associated with colon carcinogenesis. Expression is also evident in connective tissue fibroblasts. When these mice are bred with mice containing loxP-flanked sequence, Cre-mediated recombination will result in deletion of the floxed sequences in the Cre-expressing cells of the offspring. Mice express Cre in the intestinal epithelial lining during embryonic development. Heterozygotes express Cre in intestinal and colonic epithelial cells.

The donating investigator reports that the EGFP is not functional and cannot be detected by direct fluorescence or by immunofluorescence.

Development

A targeting construct was designed to replace exon 1 of the transcription factor 7 like 2, T cell specific, HMG box (Tcf7l2) gene with an enhanced green fluorescent protein fused to a Cre recombinase gene (EGFP/cre). This fusion protein sequence was followed by a frt-flanked neomycin resistance (neo) cassette. The construct was electroporated into (129X1/SvJ x 129S1/Sv)F1-Kitl⁺-derived R1-45 embryonic stem (ES) cells and correctly targeted ES cells were injected into blastocysts. The resulting Tcf4^Cre chimeric mice were bred to ACTB:FLPe mice (Stock No. 005703) to delete the neo cassette, and mice were subsequently bred to C57BL/6J mice (Stock No. 000664) for 17 generations. Upon arrival at The Jackson Laboratory, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes.

Molecular note: An EGFP/cre fusion and an FRT-flanked neo cassette replaced exon 1. Flp-mediated recombination removed the neo cassette. The absence of protein expression was confirmed by immunohistochemistry on E16.5 small intestine and colon samples.
