Detailed description

TβRI^f/f floxed mice possess loxP sites flanking exon 3 of the transforming growth factor, beta receptor I (Tgfbr1) gene. TβRI is a transmembrane serine/threonine kinase that forms a heteromeric complex with TβRII. TGF-β signaling through this complex is important for the regulation of vascular development, hematopoiesis, autoimmune diseases, and chronic inflammation. Mice that are homozygous for this allele are viable and fertile. When bred to mice that express tissue-specific Cre recombinase, resulting offspring will have exon 3 deleted in the cre-expressing tissues.

Development

A targeting vector was designed by Dr. Stefan Karlsson (Lund University) to insert a loxP-flanked neomycin resistance (neo) cassette site upstream of exon 3, and a third loxP site downstream of exon 3 of the transforming growth factor, beta receptor I (Tgfbr1) gene. The construct was electroporated into 129-derived embryonic stem (ES) cells. Correctly targeted ES cells were transiently transfected with a pIC-Cre expression plasmid to delete the neo cassette. Resulting ES cells contained multiple gene rearrangements; intact floxed-exon 3, intact floxed-neo cassette, or excision of both exon 3 and the neo cassette. Correctly targeted ES cells, containing only the floxed-exon 3, were injected into C57BL/6 blastocysts. Resulting TβRI^f/f offspring were bred to CD-1 mice and C57BL/6 mice and were maintained on a STOCK background. Upon arrival, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes (Stock No. 000664).
