The Cck-eGFP line 2 BAC transgenic mouse line has the endogenous mouse cholecystokinin promoter/enhancer regions directing fluorescent reporter expression. These mice may be useful in studying CCK+ GABAergic interneurons in behavioral and molecular processes, as well as cell-type specific GABAergic disturbances in manifestations of schizophrenia.

Cck-eGFP line 2 BAC transgenic mice have eGFP fluorescent protein expression directed by the mouse cholecystokinin promoter/enhancer regions on the BAC.
The morphology and distribution of the labeled interneurons across the different brain structures show that the transgene is specifically expressed in the cell sub-populations that normally express the Cck gene; including GABAergic interneuron basket cells (primarily synapse onto cell soma) or double bouquet cells (target dendrites in the amygdala, cortex, hippocampus, hypothalamus and thalamus).
While fluorescence is faint, strong eGFP expression via immunohistochemistry is reported specifically in CCK+ cells.
Hemizygous mice are viable and fertile with no reported behavioral abnormalities. To date (December 2015), it has not been attempted to make this Cck-eGFP line 2 BAC transgenic strain homozygous. However, the donating investigator reports that the parental strain (Cck-eGFP-miGAD67 line 2 BAC transgenic mice; Stock No. 028592) is viable and fertile as homozygotes.
The Cck-eGFP line 2 BAC transgenic mouse line (also called Cck-eGFP-lox line 2) was created by Dr. Karoly Mirnics (Vanderbilt University) by breeding Cck-eGFP-miGAD67 line 2 BAC transgenic mice (see Stock No. 028592) to mice expressing Cre recombinase in germline (C57BL/6-Tg(Zp3-cre)93Knw/J; Stock No. 003651). Offspring with germline deletion of the miRNA:Gad1-containing β-globin minigene were identified (Cck-eGFP), and then bred to C57BL/6NHsd to remove the Zp3-Cre transgene. The Cck-eGFP colony was backcrossed to C57BL/6NHsd for a total of two generations, and then sent to The Jackson Laboratory in 2012.
Upon arrival, sperm was cryopreserved. If generating a live colony, an aliquot of frozen sperm may be used to fertilize C57BL/6NJ oocytes (Stock No. 005304).

When maintaining a live colony, hemizygous mice may be bred together, to wildtype (noncarrier) mice from the colony, or to C57BL/6NJ inbred mice (Stock No. 005304).
