RTIbacGFP transgenic mice have the rat podoplanin (Pdpn) promoter/enhancer sequences on a BAC transgene driving expression of EFGP. These mice may be useful for identification, tracking, and isolation of alveolar epithelial type I cells.

RTIbacGFP transgenic mice have the rat podoplanin (Pdpn) promoter/enhancer sequences driving expression of EGFP. Homozygotes are viable, fertile. Under control of Pdpn, EGFP labels all alveolar epithelial type I cells in the lung. Pulmonary expression in these mice is more restricted than endogenous mouse podoplanin. RTIbacGFP mice express levels of Pdpn similar to the amount seen in normal rat lung.
A rat Bacterial artificial chromosome (BAC) library was used to obtain a BAC (clone address 240G15) containing the entire rat podoplanin (Pdpn) gene. This BAC was modified to insert an internal ribosome entry site (IRES) fused to an enhanced green fluorescent protein (EGFP) sequence into the 3’ UTR of the Pdpn gene. This RTIbacGFP transgene was microinjected into fertilized C57BL/6xDBA2 F1 oocytes and hemizygous mice from founder line 16 were bred to C57BL/6 mice to establish a colony. RTIbacGFP transgenic mice were bred to C57BL/6NCrl mice. Upon arrival at The Jackson Laboratory, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6NJ oocytes (Stock No. 005304).

Molecular Note | RTIbac clone 240G15 contains an insert of 168 kb inserted in the pBeloBAC11 vector. The sequence corresponds to nucleotides 162144322 to 162282008 of rat chromosome 5 and contains the rat podoplanin gene. An IRES-GFP sequence was placed in the 3'UTR. The resulting transgene directs expression of both rat podoplanin and GFP. Two stable founders were identified showing similar expression levels. One line was used for further study. |

To ensure that JAX® Mice will meet the needs of individual research projects or when requesting a strain that is new to your research, we suggest ordering and performing tests on a small number of mice to determine suitability for your particular project. We do not guarantee breeding performance and therefore suggest that investigators order more than one breeding pair to avoid delays in their research.
