Cdk1L/L floxed mice possess loxP sites flanking exon 3 of the cyclin-dependent kinase 1. This strain may be useful for studying the regulation of mitosis in terminally differentiating cells.

Cdk1L/L floxed mice possess loxP sites flanking exon 3 of the cyclin-dependent kinase 1 (Cdk1) gene. CDK1 encodes a serine/threonine protein kinase protein that is a catalytic subunit of the protein kinase complex known as M-phase promoting factor (MPF), which is essential for G1/S and G2/M phase transitions of eukaryotic cell cycle. Mice that are homozygous for this allele are viable and fertile. When bred to mice that express tissue-specific Cre recombinase, resulting offspring will have exon 3 deleted in the cre-expressing tissues.
The donating investigator reports that when bred to mice carrying the Tg(Hspa2-cre)1Eddy transgene (Stock No. 008870), expressing Cre in spermatocyte germ cells, the CDK1-null spermatocytes undergo synapsis, chiasmata formation and desynapsis, but fail to form condensed bivalent chromosomes despite initiation of chromosome condensation, and arrested at the end of prophase of meiosis I. This results in the absence of post-meiotic germ cells and male infertility.
A targeting vector was designed to insert a loxP site upstream of exon 3 followed by a frt-flanked neomycin resistance (neo) cassette, and a second loxP site downstream of exon 3 of the cyclin-dependent kinase 1 (Cdk1) gene. The construct was electroporated into 129S6/SvEvTac-derived TC-1 embryonic stem (ES) cells. Correctly targeted ES cells were injected into C57BL/6NCrl blastocysts and resulting chimeric males were bred with C57BL/6NCrl females. Resulting Cdk1L/L mice were bred to
129S6/SvEvTac mice for at least 12 generations. Upon arrival, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize 129S1/SvImJ oocytes (Stock No. 002448).
