This Maf1 ZFN (zinc finger nuclease) targeted mutation strain exhibits resistance to diet induced obesity. These mice may be suitable for use in studies related to metabolic homeostasis and obesity.

MAF1 homolog (S. cerevisiae) responds to cellular nutrient limitation and cell stress by repressing RNA polymerase III transcription, thus regulating metabolic efficiency.
These mice carry a zinc finger nuclease (ZFN) mediated single base pair insertion in the Maf1 gene causing early translational termination (shortly after Thr64). Homozygotes exhibit a phenotype similar to the phenotype observed in mice homozygous for a null allele. Mutant mice are resistant to diet induced obesity. Mice that are homozygous for the targeted mutation are viable and fertile, but may produce fewer pups and have fewer birth events.
Zinc finger nuclease (ZFN) editing induced a single base pair mutation in the Maf1 gene. The resulting change in reading frame causes a termination of translation after Thr64. ZFN mRNA was injected into C57BL/6J oocytes. Founder mice were maintained on the C57BL/6J background. Upon arrival, sperm was cryopreserved. To establish a live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes (Stock No. 000664).

When maintaining a live colony, these mice can be bred as homozygotes. However, homozygotes may exhibit reduced
fertility and fecundity (may produce fewer pups and have fewer birth events).
