Tag-Trpv1 mice may be useful for identifying modulators of pain hypersensitivity signaling through the TRPV1 receptor.

Tag-Trpv1 mice express a wild-type flag-strep-tagged mouse transient receptor potential cation channel, subfamily V, member 1 (Trpv1) gene directed by its endogenous promoter/enhancer regions on the BAC transgene. These mice overexpress the flag-strep-tagged wildtype mouse TRPV1 protein in dorsal root ganglia. TRPV1 is a non-selective cation channel involved in mediation of inflammatory pain and hyperalgesia. It is activated by temperature greater than 43 °C (109 °F), acidic conditions, and capsaicin. Hemizygotes are viable and fertile. The donating investigator reports that the Tag does not impair trafficking of TRPV1 to peripheral and central nerve terminals. Mice display increased sensitivity to pain stimuli under basal conditions.
Bacterial artificial chromosome (BAC) library (CHORI) was used to obtain a BAC (#RP23-390G23) containing the entire mouse Trpv1 (transient receptor potential cation channel, subfamily V, member 1) gene. This BAC, containing a Strep-Flag tag fused to the N terminus and in frame with the start codon of Trpv1, was microinjected into fertilized C57BL/6 oocytes. Tag-Trpv1 founder line 5917 was established and then subsequently maintained by breeding transgenic mice with C57BL/6 inbred mice, with at least the last 6 generations being backcrossed to C57BL/6NCrl mice. Upon arrival at The Jackson Laboratory, mice were bred to C57BL/6NJ inbred mice (Stock No. 005304) for at least one generation to establish the colony.

When maintaining a live colony hemizygous mice may be bred to wildtype (noncarrier) mice from the colony or to C57BL/6NJ (Stock No. 005304). The donating investigator has never tried to make the strain homozygous.
