These transgenic mice express a soluble form of chicken ovalbumin in the epidermis (keratinocytes) where it serves as a model self-antigen. K14-mOVA transgenic mice may be useful in applications related to graft-versus-host disease and peripheral immune tolerance.

These K14-sOVA transgenic mice express a soluble ovalbumin
(sOVA) protein under the direction of the human KRT14, keratin 14, gene promoter.
Transgene expression (mRNA) levels in this K14-sOVA(15) line are higher than ovalbumin transgene mRNA levels detected in the K14-mOVA (membrane-bound ovalbumin (mOVA) fusion protein) transgenic line (Stock No. 026560).
Transgenic mice are tolerant to ovalbumin antigen and do not exhibit delayed-type hypersensitivity when primed and challenged with ovalbumin.
After intradermal injection of CD8+ T cells isolated from OT-I transgenic mice (see, for example, Stock No. 003831), K14-sOVA(15) transgenic mice lose weight and develop graft-versus-host
disease-like skin and esophageal lesions, and die 4 to 5 days after adoptive transfer. The response observed in this strain is more severe than that seen in the K14-mOVA (membrane-bound ovalbumin (mOVA) fusion protein) transgenic line (Stock No. 026560).
The Donating Investigator reports that attempts to make the strain homozygous for the transgene were unsuccessful.
A transgenic construct containing a soluble ovalbumin (sOVA) protein, the 100-1167 fragment, under the control of the 2.1kb human KRT14, keratin 14, gene promoter, was injected into fertilized C57BL/6 mouse eggs.
Founder line 15 was subsequently established (see SNP note below). Upon arrival at The Jackson Laboratory, the mice were crossed to C57BL/6J (Stock No. 000664) at least once to establish the colony.
A 32 SNP (single nucleotide polymorphism) panel analysis, with 27 markers covering all 19 chromosomes and the X chromosome, as well as 5 markers that distinguish between the C57BL/6J and C57BL/6N substrains, was performed on the rederived living colony at The Jackson Laboratory Repository. While the 27 markers throughout the genome suggested a C57BL/6 genetic background, all 5 markers that determine C57BL/6J from C57BL/6N were found to be segregating. These data suggest the mice sent to The Jackson Laboratory Repository were on a C57BL/6N genetic background.

When maintaining a live colony, hemizygous mice may be bred together, to wildtype siblings, or to C57BL/6J inbred mice (Stock No. 000664). The Donating Investigator reports that attempts to make the strain homozygous for the transgene were unsuccessful.

To ensure that JAX® Mice will meet the needs of individual research projects or when requesting a strain that is new to your research, we suggest ordering and performing tests on a small number of mice to determine suitability for your particular project. We do not guarantee breeding performance and therefore suggest that investigators order more than one breeding pair to avoid delays in their research.
