Description

These transgenic mice express a membrane-bound form of chicken ovalbumin in the epidermis (keratinocytes) where it serves as a model self-antigen. K14-mOVA transgenic mice may be useful in applications related to graft-versus-host disease and peripheral immune tolerance.

Detailed description

These K14-mOVA transgenic mice express a membrane-bound ovalbumin (mOVA) fusion protein under the direction of the human KRT14, keratin 14, gene promoter. Transgene expression (mRNA) is detected by RT-PCR analysis in skin, but not in other tissues (brain, lymph node, heart, liver, spleen, kidney, intestine, skeletal muscle, esophagus). Transgenic mice are tolerant to ovalbumin antigen and do not exhibit delayed-type hypersensitivity when primed and challenged with ovalbumin. After intravenous or intradermal injection of CD8+ T cells isolated from OT-I transgenic mice (see, for example, Stock No. 003831), K14-mOVA transgenic mice lose weight and develop graft-versus-host disease-like skin and esophageal lesions. Lymphocytes from ovalbumin-immunized transgenic mice have reduced proliferative response to ovalbumin. Mice that are homozygous for the transgene are viable and fertile.

Development

A transgenic construct containing a membrane-bound ovalbumin(mOVA) fusion protein construct under the control of the 2.1kb human KRT14, keratin 14, gene promoter, was injected into fertilized C57BL/6 mouse eggs. The membrane-bound ovalbumin (mOVA) fusion protein construct consists of the murine immunoglobulin k chain V-J2-C signal peptide, a truncated ovalbumin cDNA fragment (200–1167), and sequence encoding the transmembrane domain of platelet-derived growth factor receptor (PDGFR). Founder line 6m (6-mOVA) was subsequently established. Upon arrival at The Jackson Laboratory, the mice were crossed to C57BL/6J (Stock No. 000664) at least once to establish the colony.

A 32 SNP (single nucleotide polymorphism) panel analysis, with 27 markers covering all 19 chromosomes and the X chromosome, as well as 5 markers that distinguish between the C57BL/6J and C57BL/6N substrains, was performed on the rederived living colony at The Jackson Laboratory Repository. While the 27 markers throughout the genome suggested a C57BL/6 genetic background, all 5 markers that determine C57BL/6J from C57BL/6N were found to be segregating. These data suggest the mice sent to The Jackson Laboratory Repository were on a C57BL/6N genetic background.
