The ACB017 embryonic stem cell (ESC) line was established from a cross between a B6C3-Tg(APPswe,PSEN1dE9)85Dbo/Mmjax (APP/PS1) hemizygous female (MMRRC#034829) and a C57BL/6 congenic male homozygous for Mapttm1(EGFP)Klt. This cell lines may be useful in in vitro experiments directed towards differentiation of ESCs to cortical neurons expressing AD-relevant mutations.

The ACB017 embryonic stem cell (ESC) line was established using day 3.5 blastocysts from a cross between a B6C3-Tg(APPswe,PSEN1dE9)85Dbo/Mmjax hemizygous female (MMRRC #034829/JAX#004462) and a C57BL/6 congenic male homozygous for Mapttm1(EGFP)Klt. This cross also resulted in biological replicates that are hemizygous for Tg(APPswe,PSEN1dE9)85Dbo (MMRRC#037549, #037550), as well as wild type controls (MMRRC#037546, #037547); all lines are heterozygous for EGFP fused Mapt (Mapttm1(EGFP)Klt) and wild-type for Tg(MAPT)8cPdav. These cell lines have been proven useful in in vitro experiments directed towards differentiation of ESCs to cortical neurons. The APPswe and PSEN1dE9 transgenes are also available as ESC lines generated from various crosses (see MMRRC#037551-#037553 and #037564-#037568). Cell lines were derived and maintained on mitomycin-C treated mouse embryonic fibroblast feeder cells (MEFs) derived from C57BL/6J embryos. This cell line was not germline tested.

The Jackson Laboratory has rigorous genetic quality control and mutant gene genotyping programs to ensure the genetic background of JAX® Mice strains as well as the genotypes of strains with identified molecular mutations.
