These mice express Cre recombinase under the control of a truncated follicle stimulating hormone receptor (Fshr) promoter. FSHR is a G-protein coupled receptor involved in gonad development. Hemizygotes are viable and fertile. When crossed with a strain containing a loxP site-flanked sequence, Cre-mediated recombination results in deletion of the flanked sequence in ovarian granulosa cells and in a subset of cells from the anterior pituitary. FSHR promoter activity is also seen in mammary epithelium and in the olfactory bulb. The donating investigator maintains this colony by breeding wildtype females to hemizygous males to prevent germ line deletion since Cre is produced in ovarian granulosa cells.
When bred to a strain expressing a floxed-Brca1 gene, FSHR-deficient mice develop cysts on the ovary oviducts, and uterine horns; they also show increased estradiol biosynthesis and prolongation of the preovulatory phase (proestrus) of their estrus cycle resulting in increased estrogen exposure and ensuing consequences on estrogen responsive organs such as long bones.
A transgenic construct containing cre coding sequence under the control of a truncated follicle stimulating hormone receptor (Fshr) promoter was introduced into B6D2F1 donor oocytes. Resulting offspring were bred together and were maintained on a mixed background. Upon arrival at The Jackson Laboratory, mice were bred to C57BL/6J (Stock No. 000664) for at least one generation to establish the colony.
