Description

These Osterix-Cherry mice express a Cherry protein from the Osterix promoter regions. They may be useful for tracking Osterix expression tissues during osteoblast and chondrocyte development.

Detailed description

Mice hemizygous for the BAC Osterix-Cherry transgene are viable and fertile. These mice express a red fluorescent protein (Cherry) sequence from the wild-type mouse Sp7 (Sp7 transcription factor 7) promoter regions on the BAC transgene. Osterix is a zinc finger containing transcription factor, which contains two translation start sites and is involved in osteoblast differentiation. The Cherry fluorophore was inserted upstream of the second translation start site in exon 2 in order to detect both Osterix isoforms. At E13.5 these Osterix-Cherry mice express Cherry in craniofacial bones and weakly in the fore limbs. By E15.5 expression can be detected in the ribs and hind limbs. Osterix reporter expression is detected in the vertebrae and developing digits E17.5. Mature osteoblasts and osteocytes show Cherry expression, and expression is also seen in cells of osteoblast lineage in femur tissue sections from 10 days to 12 weeks of age. The donating investigator reports no malocclussions.

Development

Bacterial artificial chromosome (BAC) library (CHORI) was used to obtain a 39 kb BAC (#RP24-362M3) containing the entire mouse Sp7 (Sp7 transcription factor 7) gene. This BAC was modified to contain a red fluorescent protein (Cherry) sequence upstream of the second translation start site. This BAC was microinjected into fertilized CD-1 oocytes. Osterix-Cherry mice from founder line 2 were maintained by breeding transgenic mice with CD-1 mice for at least 10 generations. Upon arrival at The Jackson Laboratory, mice were bred to C57BL/6J inbred mice (Stock No. 000664) for at least one generation to establish the colony.

Molecular note: Bacterial artificial chromosome (BAC) library (CHORI) was used to obtain a 39 kb BAC (#RP24-362M3) containing the entire mouse Sp7 gene. This BAC was modified to fuse the first 17 amino acids of Sp7 to the N-terminal of the red fluorescent protein (Cherry) sequence upstream of the second translation start site. This transgene contains two translational start sites; the first is located in exon 1b and results in a long protein isoform containing 428 amino acids. The second start site is in-frame with upstream start site located at the beginning of exon 2 and results in a short protein isoform containing 410 amino acids. The Cherry reporter can be used to detect both isoforms. Strong reporter expression is observed in mature osteoblasts and osteocytes; weak expression is observed in bone marrow.
