SLA2 KO mice have a puromycin resistance cassette replacing exons 2-4 of the Src-like-adaptor 2 (Sla2) gene, abolishing gene function. SLA2 is an adapter protein expressed in a variety of hematopoietic cell types, including B cells, mast cells, macrophages, and early progenitor cells. SLA2 negatively regulates T-cell receptor (TCR) signaling and granulocyte-macrophage colony-stimulating factor (GM-CSF)-mediated inflammation. It is also important for the generation of functionally mature monocyte-derived dendritic cells (DCs). Homozygotes are viable and fertile. SLA2 KO mice do not have an overt phenotype. When bred with B6.129-Slatm1Weis/J mice (Stock No. 007174), double KO mice have dendritic cell abnormalities.
A targeting vector was designed to replace exons 2-4 of the Src-like-adaptor 2 (Sla2) gene with a puromycin resistance (puro) cassette. The construct was electroporated into 129 embryonic stem (ES) cells. Correctly targeted ES cells were injected into C57BL/6 blastocysts and the resulting chimeric males were bred to C57BL/6 females. These mice were backcrossed for at least 10 generations to C57BL/6J background. Sla2 resides on Chromosome 2 in close proximity to the agouti coat color locus. The donating investigator reports that, despite extensive backcrossing to C57BL/6, the mice remain agouti. Upon arrival at The Jackson Laboratory, mice were bred to C57BL/6J (Stock No. 000664) for at least one generation to establish the colony.
