These Dlx5/6-Cre-IRES-EGFP transgenic mice may be useful for generating specific deletions of floxed alleles in ventral forebrain neuronal populations with fluorescent expression.

Hemizygous Dlx5/6-Cre-IRES-EGFP (CIE) transgenic mice are viable and fertile. Expression of Cre recombinase (Cre) and EGFP fluorescence is directed to differentiating and migrating ventral forebrain neurons, including GABAergic neurons, during embryonic development by the mouse id6/id5 enhancer. When Dlx5/6-CIE transgenic mice are bred with mice containing a loxP-flanked sequence, Cre-mediated recombination will result in deletion of the floxed sequence in the vast majority of ventral forebrain neuronal populations of the offspring.
The Dlx5/6-cre-IRES-EGFP transgene was designed with a 0.5 kb fragment containing the id6/id5 enhancer and a beta-globin minimal enhancer placed immediately upstream of a Cre recombinase coding sequence and an internal ribosomal entry site (IRES)-enhanced green fluorescent fusion protein. The construct was microinjected into FVB/N fertilized oocytes, and founder mice were initially bred to C57BL/6 mice to establish a colony. Resulting offspring were bred to CD-1 mice for at least 10 generations. Upon arrival at The Jackson Laboratory Repository, transgenic animals were bred with C57BL/6J mice (Stock No. 000664) for at least one generation to establish the colony.
