In this transgenic strain, expression of flag tagged mouse Wnt5a (wingless-related MMTV integration site 5A) transgene is regulated by a tetracycline operator (tetO). When mated to a mutant strain expressing tetracycline-controlled transactivator protein (tTA), expression of the Flag-tagged WNT5A protein may be regulated with the tetracycline analog doxycycline (dox) in the double mutant offspring. This strain may be useful in studies of the role of Wnt5a in early development and WNT signaling pathways.

The Wnt5a (wingless-related MMTV integration site 5A) gene encodes a protein that is a ligand for members of the frizzled family of seven transmembrane receptors and is able to inhibit Wnt/
beta-catenin signaling as well as activate alternative non-beta-catenin dependent signaling pathways. Mice hemizygous for the tetO-Flag-Wnt5A transgenic insert are viable and fertile. Expression of the Flag tagged mouse Wnt5a (wingless-related MMTV integration site 5A) transgene is regulated by a tetracycline operator (tetO); also called tetracycline-responsive element (TRE, TetRE) or tet-operator). The Donating Investigator has not attempted to make the strain homozygous. When mated to a mutant strain expressing tetracycline-controlled transactivator protein (tTA), expression of flag tagged WNT5A protein may be regulated with tetracycline or its analog doxycycline (dox) in the double mutant offspring. The Flag-tag allows for purification of bioactive tagged WNT5A and does not appear to interfere with WNT5A protein function. When crossed to B6.Cg-Gt(ROSA)26Sortm1(rtTA*M2)Jae/J mice (Stock No. 006965), transgene expression in the double mutant mice is widespread in the presence of doxycycline, but undetectable in the absence of doxycycline.
The tetO-Flag-Wnt5A transgenic insert designed by Drs. Christophe Fuerer and Renee van Amerongen (while at Stanford University with Dr. Roel Nusse) containing a Flag-tagged mouse Wnt5a (wingless-related MMTV integration site 5A) gene, under the control of the tetO, tetracycline-responsive regulatory element, was injected into fertilized FVB/N mouse eggs. Founder line F5A-17 was subsequently established. The mice were maintained on a FVB background. Upon arrival at The Jackson Laboratory, the mice were crossed to FVB/NJ (Stock No. 001800) at least once to establish the colony.
