Trp53cor1 (tumor protein p53 pathway corepressor 1; also known as LincRNA-p21) is a Trp53-regulated long intergenic non-coding RNA (lincRNA) expressed in the Cdkn1a (p21) locus on mouse Chromosome 17.
Floxed mutant animals were generated by flanking the promoter region and exon 1 of LincRNA-p21 with loxP sites. In the floxed configuration, expression is not affected. Upon Cre excision of the floxed region, expression is prevented. Homozygous floxed mice are viable and fertile and do not exhibit any overt phenotypes. This strain is useful in studies of cell cycle regulation, apoptosis and cancer.
The promoter region and first exon were flanked by loxP sites and an FRT-Neo-FRT drug selection cassette was placed downstream of exon 1, just inside the floxed region. The mutation was created in (C57BL/6 x 129S4/SvJae)F1-derived v6.5 embryonic stem (ES) cells. The neomycin resistance cassette was subsequently removed by breeding founders to a C57BL/6 background beta-actin FlpO strain. This strain was maintained on a mixed C57BL/6-129S4 genetic background by the donating laboratory.
