These transgenic mice carry the Brainbow 1.0 cassette, incorporating dTomato, mCerulean, and EYFP, driven by the broadly-expressed CAG (CMV-chicken beta actin) promoter. These fluorescent markers are useful in tracing cell lineage.
This strain, line 2, carries at least 5 copies of the transgene. dTomato red fluorescent protein is expressed strongly in a variety of tissues and developmental stages. Exposure of the transgene to Cre recombinase stochastically alters individual copies of the transgenes such that either EYFP (enhanced yellow fluorescent protein) or mCerulean (blue fluorescent protein) become expressed, resulting in labeling of cells with distinct colors. As many as 21 distinct color combinations have been observed in this line. Expression is reportedly seen in E3.5, 4.5, 6.5, and 7.5 embryos as well as neonates. In adult mice, expression has been observed in skeletal and cardiac muscle, kidneys, lungs, brain, spinal cord, and a portion of the eye, stomach, pancreas, and intestine. By FACS, expression has been observed in thymocytes and splenocytes, as well as CD4, CD8, CD19, CD11b and CD11c positive cells. No expression has been observed in adult liver. These alterations are genetic and heritable by the progeny of the cells.
Homozygous mice show an elevated tendency for seizures and early death not seen in the hemizygotes, likely due to high expression of dTomato.
The Brainbow 1.0 cassette (encoding lox2272-flanked dTomato, lox2272/loxP-flanked mCerulean, and EYFP fluorescent markers) was placed under the control of a CAG (cytomegalovirus (CMV) early enhancer element and chicken beta-actin) promoter. Linearized transgenic vector was injected into B6D2F1 embryos. Resultant mice were backcrossed to C57BL/6 for 9 generations by the donating lab. This is founder line 2, reported to carry at least 5 copies of the transgene.

Payment for services, products, shipping containers, and shipping costs that are rendered are expected within the payment terms indicated on the invoice or stated by contract. Invoices and account balances in arrears of stated terms may result in The Jackson Laboratory pursuing collection activities including but not limited to outside agencies and court filings.
The Jackson Laboratory has rigorous genetic quality control and mutant gene genotyping programs to ensure the genetic background of JAX® Mice strains as well as the genotypes of strains with identified molecular mutations. JAX® Mice strains are only made available to researchers after meeting our standards. However, the phenotype of each strain may not be fully characterized and/or captured in the strain data sheets. Therefore, we cannot guarantee a strain's phenotype will meet all expectations. To ensure that JAX® Mice will meet the needs of individual research projects or when requesting a strain that is new to your research, we suggest ordering and performing tests on a small number of mice to determine suitability for your particular project. We do not guarantee breeding performance and therefore suggest that investigators order more than one breeding pair to avoid delays in their research.
