Mapk3 knock-out mice (Erk1-/-) are susceptible to experimental autoimmune encephalomyelitis. They may be useful for studying the role of MAPK3 in cellular development.

A neo cassette replaces the 3' end of exon 1 through exon 6 of the mitogen-activated protein kinase 3 (Mapk3) gene, abolishing gene function. MAPK3, also known as ERK1, is part of the Ras-Raf-ERK signal transduction cascade and is involved in cell growth, differentiation, apoptosis, and transformation in response to extracellular signals in most tissues. MAPK3 is also involved in early cardiac growth and protection from apoptosis. Mice that are homozygous for this allele are viable and fertile. These knockout mice are susceptible to experimental autoimmune encephalomyelitis (EAE), are hyperactive, but have normal T cell effector function.
A targeting vector was designed to replace the 3' end of exon 1 through exon 6 of the mitogen-activated protein kinase 3 (Mapk3) gene with a loxP-flanked neomycin resistance (neo) cassette in reverse orientation to the gene. The construct was electroporated into (129X1/SvJ x 129S1/Sv)F1-Kitl+-derived R1 embryonic stem (ES) cells. Correctly targeted ES cells were injected into C57BL/6J blastocysts and the resulting chimeric males were bred to CD1 females to generate a colony of Erk1-/- mice. These mice were bred to C57BL/6J mice for at least 11 generations. Upon arrival at The Jackson Laboratory, mice were bred to C57BL/6J (Stock No. 000664) for at least one generation to establish the colony.
