Exon 2 of the vacuolar protein sorting 26 homolog A (Vps26a) gene was replaced with a neo cassette, abolishing gene function. Vps26a gene product, Hβ58, is expressed in the hippocampal formation as part of a large multimeric complex called the retromer complex. This complex is involved in protein trafficking from endosomes to the trans-Golgi network. Homozygotes display delayed development beginning at E7.5 and embryonic arrest at about E9.5. Heterozygous mice are viable and fertile, and develop hippocampal-dependent memory and synaptic dysfunction associated with elevations concentrations of Aβ peptide.
A targeting vector was designed to replace exon 2 of the vacuolar protein sorting 26 homolog A (Vps26a) gene with a neomycin(neo) resistance cassette and a polyadenylation sequence. The construct was electroporated into 129S/SvEv-Gpi1c-derived CCE embryonic stem (ES) cells. Correctly targeted ES cells were injected into C57BL/6J blastocysts and the resulting chimeric males were bred to C57BL/6J females. These mice were backcrossed to 129/SvEvTac mice for at least 5 generations. Upon arrival at The Jackson Laboratory, mice were bred to 129S1/SvImJ mice (Stock No. 002448) for at least one generation to establish the colony.

When maintaining a live colony, heterozygous mice may be bred together or to wildtype mice from the colony. The donating investigator reports that homozygotes do not survive past E9.5 due to developmental arrest.
