These Meox1-nucTagRFP-T-IRES-CE transgenic mice utilize a tamoxifen inducible Cre-mediated recombination system and may be useful in studies of genitourinary system development.

These Meox1-nucTagRFP-T-IRES-CE mutant mice have a tamoxifen inducible Cre-mediated recombination system and express the nuclear-localized Tag-RFPT variant of Red Fluorescent Protein under the direction of the mouse Meox1, mesenchyme homeobox 1, promoter. Transgenic RFP expression is not detected in wholemount embryos by fluorescence microscopy or immunohistochemistry. Tamoxifen inducible Cre recombinase activity is detected in Six2 (sine oculis-related homeobox 2 homolog (Drosophila) ) positive cap mesenchyme cells in the developing nephron at 15.5dpc. Mice that are hemizygous for the knock in allele are viable, and fertile.
This strain was transferred from the collection of the GenitoUrinary Development Molecular Anatomy Project (GUDMAP).
A targeting construct containing a nuclear-localized Tag-RFPT variant of Red Fluorescent Protein, cre/ERT2 and FRT site flanked Kanamycin/neomycin selection cassette was inserted into the consensus start ATG codon of the mouse Meox1 gene in the RP23-5O22 BAC. The PGK-Neo/Kan cassette was removed by transient Flpe-recombinase expression. The resulting modified nucTagRFPT-ires-CE BAC transgene was microinjected into (C57BL/6 x DBA) F1 donor eggs. Founder line 30 was subsequently established. The mice were then crossed to C57BL/6 for 3 generations. Upon arrival at The Jackson Laboratory, the mice were crossed to C57BL/6J (Stock No. 000664) at least once to establish the colony.
