This strain has been discontinued. The Jackson Laboratory quality control efforts have confirmed very low mCherry intensity in this strain. A suitable alternative may be the Flk1-myr::mCherry transgenic line (Stock No. 032145) or Flk1-GFP knock-in/knock-out line (Stock No. 017006). Flk1-myr::mCherry transgenic mice express a myristoylated mCherry fluorescent protein under control of Kdr regulatory regions specifically in endothelial cells and have applications in in vivo studies of blood vessels and vasculature and for breeding multi-fluorescent protein expressing mutant lines.

In these transgenic mice mCherry fluorescent protein expression is specific to endothelial cells (myristoylation targets the mCherry expression to the cell membrane). Fluorescence is detected in embryonic endothelium and endocardium as early as 8.5 dpc, in ocular vessels and in capillaries of adult animals. Mice that are homozygous for the transgene are viable, fertile and have a higher transgene expression level than hemizygotes, resulting in brighter fluorescence.
Researchers using this strain have notified The Jackson Laboratory that the mCherry intensity in this mouse line is very low, and may not accurately reflect Kdr expression. Jackson Laboratory quality control efforts have confirmed these observations. Stock No. 017006 and Stock No. 032145 may be suitable alternatives.
In this transgenic construct a mCherry fluorescent protein gene fused to a myristoylation
motif and SV40 polyadenylation site sequence was inserted between the mouse Kdr, kinase insert domain protein receptor, regulatory elements (promoter and enhancer). This construct was injected into fertilized FVB mouse eggs. Founder line 1 was subsequently established. The strain was maintained on the FVB background by the Donating Investigator. Upon arrival at The Jackson Laboratory, the mice were crossed to FVB/NJ (Stock No. 001800) at least once to establish the colony.
