These Krt17cre/cerulean mice express cre recombinase and the cerulean variant of green fluorescent protein (GFP) driven by the endogenous keratin 17 (Krt17) promoter/enhancer elements. They may be useful for generating conditional mutations for studying craniofacial development.

Mice heterozygous for the Krt17cre/cerulean allele expresses cre recombinase and the cerulean variant of green fluorescent protein (GFP) from endogenous keratin 17 (Krt17) promoter/enhancer elements. Craniofacial-specific Krt17-cre activity has been detected, starting at E13.5, in tooth bud, hair follicle, and regions of nasal, oral, and eyelid epithelium. When these mice are bred with mice containing a loxP-flanked sequence, cre-mediated recombination will result in deletion of the floxed sequences in the cre-expressing cells of the offspring.
In the laboratory of Dr. Steve Murray at the Jackson Laboratory, a targeting vector was designed to replace the stop codon of the endogenous keratin 17 (Krt17) gene with (from 5' to 3') a viral 2A oligopeptide (to mediate ribosomal skipping), cre recombinase, another 2A oligopeptide, the cerulean variant of green fluorescent protein (GFP), and a bovine polyadenylation sequence. This sequence was followed by a frt-flanked neomycin (neo) resistance cassette. This construct was electroporated into C57BL/6NCrl derived JM8A3 embryonic stem (ES) cells. Correctly targeted ES cells were injected into recipient blastocysts and chimeric males were bred with C57BL/6NJ females to establish a colony. Upon arrival at The Jackson Laboratory Repository, mice were bred with C57BL/6NJ inbred mice (Stock No. 005304) for at least one generation.

To ensure that JAX® Mice will meet the needs of individual research projects or when requesting a strain that is new to your research, we suggest ordering and performing tests on a small number of mice to determine suitability for your particular project.
