An EGFP, followed by an SV40 polyA signal, was inserted into the ATG start site of the Slc17a8 gene within the BAC transgene. VGLUT3 is a vesicular glutamate transporter that mediates synaptic transmission by transporting the neurotransmitter, glutamate, into secretory vesicles. Mice homozygous for this allele are viable, fertile, normal in size, and do not display any gross physical or behavioral abnormalities. EGFP signal is evident in dorsal root ganglion in the dorsal horn, lamina I, lamina II, and ipsilateral dorsal horn of the spinal cord.
The C57BL/6 mouse bacterial artificial chromosome (BAC) RP24-88H21, containing the entire Slc17a8 locus only, was modified by targeting an enhanced green fluorescent protein (EGFP), followed by an SV40 polyA signal, into the ATG start site of the Slc17a8 locus. The modified BAC was microinjected into fertilized B6SJL oocytes and transgenic founders were obtained. These VGLUT3 eGFP mice were maintained on a mixed C57BL/6/SJL/DBA2 mixed genetic background. Upon arrival at The Jackson Laboratory, transgenic mice were bred with C57BL/6J inbred mice (Stock No. 000664) for at least one generation to establish this colony.

When maintaining a live colony, hemizygous mice may be bred to wildtype (noncarrier) mice from the colony or to C57BL/6 inbred mice (Stock No. 000664). The donating investigator reports that homozygous mice are viable and fertile.
