EGFP and the human SOD1 G37R mutation are driven by an inducible upstream activation sequence (UAS) promoter in this transgenic strain. These mice may be crossed with a GAL4 strain to activate gene expression in studies of familial amyotrophic lateral sclerosis (ALS) and motor neuron disease.

Enhanced green fluorescent protein (EGFP) and human SOD1 (superoxide dismutase 1, soluble) carrying the G37R mutation are each driven by the an inducible upstream activation sequence (UAS) promoter in this transgenic strain. Hemizygotes appear healthy, develop normally, and do not display any impairment of viability or reproductive capacity.
These animals express EGFP and SOD1 at very low levels. When crossed with a GAL4-expressing strain (e.g. Stock No. 017456 or Stock No. 017457 crossed with a Cre strain), EGFP and SOD1 are expressed at at higher levels. This strain may be effectively used as a component of the GAL4-UAS (Upstream Activation Sequence) system to activate gene transcription in studies of familial amyotrophic lateral sclerosis (ALS) and motor neuron disease.
A construct carrying two 380 bp upstream activation sequence (UAS) DNA fragments that act as inducible promoters was designed to express enhanced green fluorescent protein (EGFP) and human SOD1 G37R mutant sequences (UAS-EGFP-UAS-SOD1(G37R)). The vector was introduced to C57BL/6 x BALB/c hybrid animals. This strain was backcrossed to C57BL/6 once by the donating laboratory.

When maintaining the live colony, hemizygous mice may be bred with wildtype (noncarrier) mice from the colony or with C57BL/6J inbred mice (Stock No. 000664). Homozygous viability/fertility were not determined by the donating laboratory.

The Jackson Laboratory has rigorous genetic quality control and mutant gene genotyping programs to ensure the genetic background of JAX® Mice strains as well as the genotypes of strains with identified molecular mutations. JAX® Mice strains are only made available to researchers after meeting our standards. However, the phenotype of each strain may not be fully characterized and/or captured in the strain data sheets. Therefore, we cannot guarantee a strain's phenotype will meet all expectations. To ensure that JAX® Mice will meet the needs of individual research projects or when requesting a strain that is new to your research, we suggest ordering and performing tests on a small number of mice to determine suitability for your particular project. We do not guarantee breeding performance and therefore suggest that investigators order more than one breeding pair to avoid delays in their research.
