These targeted mutant mice possess loxP sites on either side of the third coding exon of the Pot1a (protection of telomeres 1A) gene. Mice that are homozygous for this floxed allele are viable, fertile, normal in size and do not display any gross physical or behavioral abnormalities. When bred to mice that express Cre recombinase, resulting offspring will not express the targeted gene in cre-expressing tissues.
A cassette containing loxP-neomycin-loxP-FRT-puromycin-STOP-FRT was introduced to the intron preceding the third coding exon of the gene. An additional loxP site was placed in the following intron. The mutation was created in E14 129P2/OlaHsd-derived embryonic stem (ES) cells. The puromycin-STOP cassette was excised through crosses with 129S4/SvJaeSor-Gt(ROSA)26Sortm1(FLP1)Dym/J transgenic mice (Stock No. 003946) mice, leaving the neomycin cassette and coding exon 3 flanked by loxP sites. This strain was maintained on a mixed C57BL/6 and 129 genetic background by the donating laboratory. Upon arrival at The Jackson Laboratory Repository, mutant mice were bred to C57BL/6J inbred mice (Stock No. 000664) for at least one generation to establish the colony.
