This strain is one of a set of 3 NOD.B6 congenic Idd10 and Idd18 strains (Stock No. 013185, 013187). Stocks 13185, 13186 and 13187 resulted from crossing segments of C57BL/6J derived chromosome 3 into the NOD/MrkTac background. Mice of this strain contain C57BL/6J derived Idd10, Idd18.1, Idd18.2 and Idd18.3 regions. These strains may be useful to better understand the variation of diabetic resistance conferred by Idd loci; for studying long-range chromosome remodeling and may be useful for identifying diabetes susceptible candidate genes within the Idd18 subloci.

This NOD/MrkTac congenic strain, commonly referred to as NOD.B6-Idd10/18, line 7754 is viable and fertile. Mice of this strain are derived from line 1101 and contain a genomic segment from the diabetes resistant strain C57BL/6J (Stock No. 000664) introgressed into the NOD chromosome 3. The C57BL/6 genomic segment includes Idd10, Idd18.1, Idd18.2 and Idd18.3; , while the Idd3 and Idd17 regions are of NOD origin. The Idd10 region contains major candidate gene Cd101 (100Mb); the Idd18.1 region contains major candidate gene Vav3 and the recently identified 966Kb Idd18.3 region contains 22 genes and 1 pseudogene.
Thymocytes of 3-week old male congenic mice express 1.46 fold less full length VAV3 mRNA than thymocytes of line 2412 (Stock No. 013187); which carries a C57BL/6J derived Idd10, Idd18.2 and Idd18.3. Diabetes incidence overlaps with line 2412 and is reported to be about 55%.
This strain is useful for understanding the variation in diabetic resistance conferred by Idd loci, in particular the epistatic interactions of Idd3 alleles with alleles at Idd18 and for conducting mapping experiments to further define the Idd18.3 region.
This strain, NOD.B6-Idd10C57BL/6J Idd18C57BL/6J/7754MrkTacJ, was produced by mating mice from NOD/MrkTac (NOD) to C57BL/6J followed by 10 additional crosses to NOD with selection for chromosome 3, Idd10, Idd18.1, Idd18.2 and Idd18.3 markers. Markers D3Mit213 (97.98Mb) and RS38124092 (122Mb) are C57BL/6J derived, while markers D3Mit41 (97.59Mb) and D3Mit370 (125Mb) are of NOD origin. Siblings carrying the C57BL/6J derived segments were intercrossed to make homozygous. 5kSNP Chip Data, based on NCBI, build 36, identifies no C57BL/6J genetic contamination of other chromosomes. In 2010, the Type 1 Diabetes Resource received this strain at generation N11F12.

The Jackson Laboratory has rigorous genetic quality control and mutant gene genotyping programs to ensure the genetic background of JAX® Mice strains as well as the genotypes of strains with identified molecular mutations. JAX® Mice strains are only made available to researchers after meeting our standards. However, the phenotype of each strain may not be fully characterized and/or captured in the strain data sheets. Therefore, we cannot guarantee a strain's phenotype will meet all expectations. To ensure that JAX® Mice will meet the needs of individual research projects or when requesting a strain that is new to your research, we suggest ordering and performing tests on a small number of mice to determine suitability for your particular project. We do not guarantee breeding performance and therefore suggest that investigators order more than one breeding pair to avoid delays in their research.
