In this knockin strain tauGFP replaces exon 1; expression of tauGFP is controlled by Itpr3 regulatory elements and is directed to microvillous cells in the olfactory epithelium. This mutant mouse strain may be useful in studies of taste transduction.

Homozygotes: Mice that are homozygous for the targeted mutation are viable, fertile, normal in size and do not display any gross physical or behavioral abnormalities. Expression of the microtubule-associated protein tau/Green Fluorescent Protein fusion gene, "tauGFP" is controlled by Itpr3 regulatory elements. GPF expression is directed to microvillous cells in the olfactory epithelium. This mutant mouse strain may be useful in studies of taste transduction.
Heterozygote: Heterozygote phenotype is expected to be similar to homozygote phenotype.
A targeting vector containing a tau (Mapt)/GFP fusion protein followed by three polyA sites and a loxP-flanked PGKneo cassette was used to replace exon 1. The construct was electroporated into (C57BL/6 x 129X1/SvJ)F1 derived EC7.1 embryonic stem (ES) cells. The resulting chimeric animals were crossed to FVB/N-Tg(ACTB-cre)2Mrt/J mice to remove the neomycin cassette. The strain then was crossed to C57BL/6 mice for two generations. The cre transgene was bred out of this strain. Upon arrival, mice were bred to C57BL/6J for at least 1 generation to establish the colony.

The Jackson Laboratory has rigorous genetic quality control and mutant gene genotyping programs to ensure the genetic background of JAX® Mice strains as well as the genotypes of strains with identified molecular mutations. JAX® Mice strains are only made available to researchers after meeting our standards. However, the phenotype of each strain may not be fully characterized and/or captured in the strain data sheets. Therefore, we cannot guarantee a strain's phenotype will meet all expectations. To ensure that JAX® Mice will meet the needs of individual research projects or when requesting a strain that is new to your research, we suggest ordering and performing tests on a small number of mice to determine suitability for your particular project. We do not guarantee breeding performance and therefore suggest that investigators order more than one breeding pair to avoid delays in their research.
