A loxP-flanked neomycin resistance cassette was inserted into intron 1, and a Ser59Asn (ATCTGC to ATTAAT) mutation was created in exon 2, introducing an Ase1 restriction site. TC1 embryonic stem (ES) cells derived from 129S6/SvEvTac were used to create the mutation. The floxed neomycin cassette was excised through a germline C57BL/6 background protamine (Prm) PC3-cre recombinase cross. This strain was backcrossed to C57BL/6 for 10 generations by the donating laboratory.
