These ROSA:LNL:tTA mice can be used to generate compound mutant mice in which the tissue specificity of a cre-transgenic line and the tetracycline or dox responsiveness of the tTA/TRE-controlled transgenes can be combined to regulate expression of the target gene.

Mice heterozygous for the ROSA:LNL:tTA conditional mutation of the Gt(ROSA)26Sor locus are viable and fertile, with a loxP-flanked STOP cassette preventing transcription of a downstream optimized/modified tetracycline-controlled transactivator protein ("mtTA"). Applying both Cre-lox and Tet-Off technologies, these ROSA:LNL:tTA mutant mice may be useful to generate compound mutant mice in which expression of a tetracycline-responsive promoter element (TRE; also called tet-operator or tetO)-driven gene of interest can be both directed to the cell types defined by the chosen Cre recombinase expression, as well as turned off by the addition of tetracycline (or its analog doxycycline (dox)).
The ROSA:LNL:tTA targeting vector was designed to insert a loxP-flanked STOP cassette (containing a neomycin resistance cassette and transcriptional STOP element) just upstream of a optimized form of the tetracycline controlled transactivator ("mtTA") into the Gt(ROSA)26Sor locus. The donating investigator reports that the construct was electroporated into 129P2/OlaHsd-derived HM-1 embryonic stem (ES) cells, correctly targeted ES cells were injected into recipient blastocysts and the resultant chimeric mice were bred to C57BL/6J mice to establish the colony. Mutant mice on a mixed genetic background (mostly C57BL/6 with some 129 or FVB/N) were sent to The Jackson Laboratory (as Stock No. 008600). Upon arrival, some mice were bred with C57BL/6J using a marker-assisted, speed congenic approach to generate this congenic strain (Stock No. 011008).

To ensure that JAX® Mice will meet the needs of individual research projects or when requesting a strain that is new to your research, we suggest ordering and performing tests on a small number of mice to determine suitability for your particular project. We do not guarantee breeding performance and therefore suggest that investigators order more than one breeding pair to avoid delays in their research.
