Description

This compound mutant strain carries targeted knockouts of the Syngr1 (P29; synaptogyrin I) and Syp (P38; synaptophysin I) genes, and floxed conditional mutations of Synpr (SP2; synaptophysin II) and Syngr3 (SG3; synaptogyrin III). This strain may be useful in studies of synaptic plasticity and the role of synaptophysins and synaptogyrins in neurological systems.

Detailed description

This compound mutant strain carries targeted knockouts of the Syngr1 (P29; synaptogyrin I) and Syp (P38; synaptophysin I) genes, and floxed conditional mutations of Synpr (SP2; synaptophysin II) and Syngr3 (SG3; synaptogyrin III). The donating laboratory reports that seizures may occur after about 2 months of age, triggered by lifting mice up by the tail (highest incidence) or other sensory stimuli (less common). This strain may be useful in studies of synaptic plasticity and the role of synaptophysins and synaptogyrins in neurological systems.

Development

This strain carries four targeted mutations, created independently then intercrossed.

Synpr (SP2) floxed conditional: LoxP sites flank exon 5. A neomycin cassette flanked by Frt sites was used for positive selection.

Syngr1 (P29) knockout: Exon 2 of this X-linked gene was replaced by a neomycin resistance cassette via homologous recombination in 129S6/SvEvTac-derived SM1 embryonic stem (ES) cells. Absence of expression was confirmed by the donating lab in total brain (Western Blot) and hippocampus (immunohistochemical).

Syngr3 (SG3) floxed conditional: LoxP sites flank exon 3, the coding portion of exon 4, and a C-terminus GFP (Venus) tag. A neomycin cassette flanked by Frt sites was used for positive selection.

Syp (P38) knockout: A genomic clone containing seven exons of the gene was used to construct a targeting vector that deletes exon 3 and most of the genomic sequence between exons 2 and 4. A centrally located neomycin resistance gene cassette was used for selection.

The genetic background of this compound mutant strain includes contributions from C57BL/6 and 129.
