These mice express the H2B-eGFP fusion gene from the endogenous Pdgfra locus. Fluorescence patterns mimick the expression pattern of the endogenous gene. Homozygotes exhibit abnormal placenta development and placenta vasculature. This mutant mouse strain may be useful in studies of cellular signaling during development and in adult mice; specifically receptor tyrosine kinases (RTK; such as Ras, MAP kinase, PI3K and those in the platelet-derived growth factor (PDGF) family).

Mice homozygous for this knock-in targeted mutation have an embryonic lethal phenotype, with half of the embryos failing to survive past embryonic day 12.5 and the remainder failing to survive beyond embryonic day 15.5. These mice express the H2B-eGFP fusion gene from the endogenous Pdgfra locus. Fluorescence is detectable at embryonic day 4.5 in polar trophectoderm cells and at embryonic day 6.5 in the extraembryonic ectoderm. Expression of H2BGFP mimick the expression pattern of the endogenous gene. Homozygotes exhibit abnormal placenta development and placenta vasculature. A targeting vector containing a human H2B histone protein and enhanced Green Fluorescent Protein fusion gene, H2B-GFP or H2B-eGFP, and a floxed PGKneo selection cassette was used to disrupt sequence of the targeted gene that encodes the signal peptide and the first two immunoglobulin
domains of the targeted gene. The endogenous Pdgfra promoter drives expression of the H2B-eGFP fusion gene. The construct was electroporated into 129S4/SvJaeSor derived AK7 embryonic stem (ES) cells. Correctly targeted ES cells were injected into recipient blastocysts. The resulting chimeric animals were backcrossed to C57BL/6 mice for 17 generations.
