Description

Similar to mice of strain NOD.B6-(D6Mit254-D6Mit339)/CarJ (Stock #4482), these mutant mice carry a C57BL/6-derived congenic interval on Chromosome 6 which contains the Nk1.1 locus but is shorter and mice have a higher incidence of diabetes.

Detailed description

Klrb1c^a (Nk1.1) homozygous mice are viable, fertile, normal in size, and do not display any gross physical or behavioral abnormalities. Nk1.1 expression in the strain has been confirmed by FACS analysis of peripheral blood cells. The C57BL/6-derived congenic interval on Chromosome 6 which contains the Nk1.1 locus is shorter in these mice than that carried in the original strain (Stock No. 004482). Diabetes incidence in this strain is higher than that reported in strain 004482 (65% in females and 15% in males by 27 weeks for this strain versus 30% in the females and 0% males by 30 weeks of age for Stock No. 004482). Histological examination of the pancreas from 27-week old, non-diabetic mice reveals extensive insulitis that is not significantly different from that seen in similarly aged Stock No. 004482 mice or NOD/ShiLtJ inbred mice (Stock No. 001976). This model is useful for looking at the role of natural killer cells in autoimmune diseases such as Type 1 Diabetes.

Development

A Chromosome 6 genomic segment from C57BL/6JOrl originally reported to span D6Mit254 through D6Mit14 and containing Klrb1c^a (commonly referred to as NK1.1) was transferred through 10 generations of backcrossing to NOD/Necker (Carnaud et al, 2001). In 2002, this strain arrived at The Jackson Laboratory at N10F? (Stock No. 004482). A partial genome scan indicated that the C57BL/JOrl congenic interval on Chr. 6 was longer than what was originally published. Stock No. 004482 was further backcrossed to NOD/ShiLtJ (Stock No. 001976) prior to making the stock homozygous for a smaller C57BL/6JOrl-derived interval (D6Mit254-D6Mit289) containing Klrb1c^a [NK1.1 (62.1 cM)]. The C57BL/6 genetic segment in this NOD congenic stock is approximately 8.5cM shorter than the original stock received. In 2007, the generation of this strain is N10+N5F4.
