These transgenic mice express a human histone H2B gene with enhanced green fluorescent protein gene under the control of the chicken beta actin promoter/enhancer (CMV). Fluorescence is detectable in nucleosomes and chromatin during all phases of mitosis in all cells. This mutant mouse strain may be useful for in vivo subcellular high-resolution and 3-dimensional studies of dividing cells.

Mice homozygous for the transgenic insert are viable, fertile, normal in size and do not display any gross physical or behavioral abnormalities. These transgenic mice express an "H2B-EGFP" fusion protein (coding sequence for the human HIST1H2BB gene [histone 1 H2bb] followed C-terminally by Enhanced Green Fluorescent Protein gene [EGFP, BD Biosciences]) under the control of the chicken beta actin promoter/enhancer coupled with the cytomegalovirus (CMV) immediate-early enhancer. Nucleosomes and chromatin in all cells fluoresce. Fluorescence is detectable during all phases of mitosis. The donating investigator reports occasional silencing of the transgene when transmitted through the female germline. This mutant mouse strain may be useful for in vivo subcellular high-resolution and 3-dimensional studies of dividing cells.
In an attempt to offer alleles on well-characterized or multiple genetic backgrounds, alleles are frequently moved to a genetic background different from that on which an allele was first characterized. It should be noted that the phenotype could vary from that originally described. We will modify the strain description if necessary as published results become available.
A transgenic construct containing an "H2B-EGFP" fusion protein (coding sequence for the human HIST1H2BB gene [histone 1 H2bb] followed C-terminally by Enhanced Green Fluorescent Protein gene [EGFP, BD Biosciences]) was placed under the control of a chicken beta actin promoter/enhancer coupled with the cytomegalovirus (CMV) immediate-early enhancer promoter. This construct was electroporated into (129X1/SvJ x 129S1/Sv)F1-derived R1 embryonic stem (ES) cells. H2B-EGFP expressing ES cells were injected into C57BL/6 blastocysts. Chimeric mice were bred to 129/Tac mice for germline transmission. Resulting founder animals were bred with outbred ICR mice prior to arrival at The Jackson Laboratory. The mice were then backcrossed on to the C57BL/6J background for at least five generations.

Molecular Note | The coding sequence of EGFP was placed C-terminally to the coding sequence of the human histone H2B gene. When placed under control of the chicken beta actin promoter/enhancer coupled with the cytomegalovirus (CMV) immediate-early enhancer/promoter, a fluorescent fusion protein (H2B-EGFP) is produced. |

When maintaining a live colony, hemizygous mice are bred wildtype siblings. For mice on the STOCK background (Stock No. 005418), the donating investigator reports occasional silencing of the transgene when transmitted through the female germ line and recommends identifying mutant offspring by the fluorescent phenotype.

The Jackson Laboratory has rigorous genetic quality control and mutant gene genotyping programs to ensure the genetic background of JAX® Mice strains as well as the genotypes of strains with identified molecular mutations. JAX® Mice strains are only made available to researchers after meeting our standards. However, the phenotype of each strain may not be fully characterized and/or captured in the strain data sheets. Therefore, we cannot guarantee a strain's phenotype will meet all expectations. To ensure that JAX® Mice will meet the needs of individual research projects or when requesting a strain that is new to your research, we suggest ordering and performing tests on a small number of mice to determine suitability for your particular project. We do not guarantee breeding performance and therefore suggest that investigators order more than one breeding pair to avoid delays in their research.
