These mice possess a disrupted human placental alkaline phosphatase (ALPP) transgene inserted into the Gt(ROSA)26Sor locus. Expression of the ALPP transgene is contingent upon Flp-mediated excision of the FRT-flanked STOP cassette placed between the ALPP peptide encoding sequence and the Gt(ROSA)26Sor promoter. This mutant strain serves as a reporter strain, with successful Flp recombinase-mediated excision indicated by ALPP activity. Mice that are homozygous for this allele are viable, fertile, normal in size and do not display any gross physical or behavioral abnormalities.
When bred to a strain with broad expression of FLP1 recombinase (see Stock No. 003946 for example), this mutant mouse strain may be useful as an indicator strain for marking most cells in the developing and adult mouse.
A FRT-flanked targeting vector containing a neomycin resistance gene driven by the mouse phosphoglycerate kinase promoter and four head-to-tail SV40 polyadenylation sites was used to disrupt the human placental alkaline phosphatase (ALPP) transgene. This FRT-disrupted ALPP transgene was then inserted into the Gt(ROSA)26Sor locus, disrupting expression of the Gt(ROSA)26Sor gene. The final construct, R26FRAPki, was electroporated into 129S4/SvJaeSor derived AK7 embryonic stem (ES) cells. Correctly targeted ES cells were injected into C57BL/6 blastocysts.

Molecular Note | An Flp-dependent alkaline phosphatase reporter locus was generated by inserting a construct containing an frt-flanked PGK-neo gene followed by sequence encoding human ALPP. The inserted construct contained a splice acceptor site at the 5' end. In addition to transcriptional and translational stop signals in the frt-flanked region, the frt-flanked neo cassette was inserted immediately downstream of the ALPP start codon to ensure a lack of ALPP activity in the absence of Flp-mediated excision. |

The resulting chimeric animals were bred to produce homozygous mice. When maintained in a live colony, this strain may be bred using homozygous matings. Expected coat color is: Black and agouti.
