These mice do not develop diabetes; The presence of the T cell receptor expressing transgene does not alter the resistance to type 1 diabetes conferred to the NOD background by homozygosity for the scid mutation. Because this strain is homozygous for the Emv30b allele, which is the absence of this proviral insertion, these mice have a much lower incidence of thymic lymphoma progression than normally found in mice on the NOD background.
This strain was generated by repeated backcrossing of Tg(TcrLCMV)327Sdz from a segregating B6;D2 background onto NOD.Cg-Prkdcscid Emv30b/Dvs. At N9 mice were sibling intercrossed to reach homozygosity for Prkdcscid and Emv30b and this strain was maintained segregating for Tg(TcrLCMV)327Sdz. Embryos were generated for cryopreserved from Tg/? Males and females at generation N9F24 so cryorecovered pups should be homozygous for Prkdcscid and Emv30b and segregating for the transgene.
