Male mice with this transgene on a segregating B6;SJL background develop multiple tumors of the mammary and salivary glands by 1 year of age, and on the FVB/N background they develop multiple mammary tumors by age 6-8 weeks; the tumor phenotype has not been characterized on the B6;FVB segregating background, but some males of the Edaraddcr-3J bearing stock developed visible mammary gland tumors and became moribund between 4 and 8 months of age (LR Donahue, personal communication). The transgene can be eliminated from the stock by crossing a female homozygous for Edaraddcr-3J with a wild-type male mouse, which will replace the transgene-bearing Y Chromosome with that of the wild-type strain.
Mice exhibitiing the crinkled 3 Jackson (Edaraddcr-3J) phenotype were found in 2000 by Harold Coombs in Dr. Wesley Beamer's colony of FVB/N-Tg(WapHRAS)69Lln Chr YSJL/J mice at The Jackson Laboratory. Edaraddcr-3J homozygous males were crossed to C57BL/6J females; these pairs and their F1 progeny were donated to Dr. Leah Rae Donahue in the Mouse Mutant Resource. Homozygous F2 male progeny of these crosses were again crossed to C57BL/6J females, and sperm from N2F2 males was cryopreserved. Mice recovered by in vitro fertilization using the frozen sperm and ova from C57BL/6J females will thus be at backcross generation N3 to C57BL/6J.
The background strain on which Edaraddcr-3J arose was then at backcross generation N14 to FVB/N. It carried, and still carries, a transgene comprising the human Harvey rat sarcoma (HRAS) oncogene homolog under control of the whey acidic protein promoter. The transgene was originally injected into a B6SJLF1 zygote, and it integrated into the Y Chromosome. It was transferred onto and maintained on the FVB/N background by crossing male mice hemizygous for the transgene with wild-type FVB/N female mice for 14 generations, after which it was maintained by crossing hemizygous males with their sisters.
