These 427-line mice express a transgene containing SV40 T antigen early region genes. They show a high incidence of brain choroid plexus tumors and are unresponsive to immunization with SV40antigen.

This 427 transgenic strain and the 419 strain C57BL/6J-Tg(SV)427Bri/J (Stock No. 003477) each contain the same SV40 T antigen early region genes in the SVMK plasmid construct, but differ in choroid plexus tumor incidence and immune response to SV40 T antigen. Strain 427 mice developed 95 tumors out of 96 mice while 419 mice developed 1 tumor out of 53 mice. Strain 427 mice are completely unresponsive to immunization with SV40 while mice mount a normal immune response to SV40 T antigen compared with non-transgenic control mice. The transgene, containing the SVMK plasmid consisting of a pSV3-ts58-derived Kpn I-Bam HI fragment, was injected into the pronuclei of C57BL/6J mouse oocytes. Mice from founder line 427 bred with C57BL/6J mice to establish the colony.

Transgenic males are sterile. When maintaining a live colony, hemizygous females may be bred to wildtype males, or to C57BL/6J male mice (Stock No. 000664). Transgenic males are sterile.
