Prx1-TLS:CHOP transgenic mice (pTC transgenic mice) express the fusion protein TLS:CHOP in the appendicular skeleton, and may have applications related to the therapeutic treatment of myxoid round cell liposarcoma.

A chromosomal translocation t(12;16)(q13;p11) that leads to a gene fusion encoding the TLS:CHOP (or FUS-CHOP) fusion protein is associated with myxoid round cell liposarcoma. These transgenic mice express the fusion protein TLS:CHOP under the direction of the rat Prrx1, paired related homeobox 1, promoter, in the appendicular skeleton. Mice hemizygous for the transgenic insert are viable, fertile and do not form tumors. The Donating Investigator has not attempted to make the strain homozygous.
When crossed to mice carrying a null allele for Trp53
(see Stock No. 002101), the double mutant mice develop sarcomas predominantly located in the appendicular skeleton and are a model for myxoid round cell liposarcoma.
A transgenic construct containing sequence encoding the fusion protein TLS:CHOP (type II) under the control of the rat Prrx1, paired related homeobox 1, promoter, chicken beta-globin domain sequence and SV40 polyadenlyation signal sequence, was microinjected into B6CBA/F1 fertilized mouse eggs. The resulting transgenic mice were crossed to B6CBA/F1 mice. Upon arrival at The Jackson Laboratory, the mice were crossed to C57BL/6J (Stock No. 000664) at least once to establish the colony.

To ensure that JAX® Mice will meet the needs of individual research projects or when requesting a strain that is new to your research, we suggest ordering and performing tests on a small number of mice to determine suitability for your particular project. We do not guarantee breeding performance and therefore suggest that investigators order more than one breeding pair to avoid delays in their research.
