Description

Mafb^p/p floxed mice have loxP sites flanking the single coding exon of the Mafb gene. This strain may be useful for studying endothelial sprouting and angiogenesis of retinal vasculature.

Detailed description

Mafb^p/p floxed mice have loxP sites flanking the single coding exon of the MAF bZIP transcription factor B (Mafb) gene. Mafb encodes a transcription factor that regulates endothelial sprouting and angiogenesis.

Mice that are homozygous for the Mafb^p/p allele are viable and fertile. When these mice are bred to mice that express Cre recombinase, resulting offspring will have the Mafb coding region deleted in Cre expressing tissues.

When these mice were crossed to Pdgfb-iCreER mice with tamoxifen-inducible endothelial-cell (EC)-specific Cre expression, retinal vasculature in the resulting Mafb^iΔEC mice show a reduction in EC sprouting, vascular area, and vessel branch formation when compared to control littermates after tamoxifen induction.

Development

A targeting vector was designed to insert a single loxP site upstream of the single coding exon of the MAF bZIP transcription factor B (Mafb) gene and a frt-flanked puromycin (puro) cassette followed by a second loxP site were inserted downstream of the coding exon. The construct was electroporated into C57BL/6NTac-derived embryonic stem (ES) cells and correctly targeted ES cells were injected into blastocysts. Resulting Mafb^p mice were subsequently bred to Flp mice on a C57BL/6 genetic background to remove the puro cassette and were subsequently bred to C57BL/6J mice for at least 3 generations by the donating laboratory. Upon arrival at The Jackson Laboratory, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes (Stock No. 000664).
