Description

miR-206^flox floxed mice have loxP sites flanking the stem-loop sequence of the Mir206 gene. This strain may be useful for studying the role of miR-206 in psychiatric disorders such as autism, schizophrenia, and Parkinson's disease.

Detailed description

miR-206^flox floxed mice have loxP sites flanking the stem-loop sequence of the microRNA 206 (Mir206) gene. Mir206 encodes a microRNA that acts by negative regulation of cell migration, cell adhesion molecule production, and cardiac and skeletal muscle development. miR-206 is enriched in the prefrontal cortex, hippocampus, and cerebellar Purkinje cells (PCs) where it controls neuronal excitability and sensorimotor gating. miR-206 and the co-expressed microRNA, miR-133b, have been associated with autism, schizophrenia, Parkinson's disease, and other psychiatric disorders. Mutations of this gene have also been implicated in skeletal muscle disorders, including amyotrophic lateral sclerosis and Duchenne muscular dystrophy.

Mice that are homozygous for the miR-206^flox allele are viable and fertile. When these mice are bred to mice that express Cre recombinase, the resulting offspring will have the entire stem-loop region of miR-206 deleted in Cre expressing tissues.

Development

A targeting vector was designed to insert a single loxP site and an frt-flanked neomycin resistance (neo) cassette upstream of the stem-loop sequence of the microRNA 206 (Mir206) gene, and a second loxP site was inserted downstream of the stem-loop sequence. The construct was electroporated into (129X1/SvJ x 129S1/Sv)F1-Kitl⁺-derived R1 embryonic stem (ES) cells and a correctly targeted ES cell was injected into C57BL/6J blastocysts. Resulting chimeric mice were bred to C57BL/6J mice (Stock No. 000664), and the miR-206^Neo-flox offspring were subsequently bred to Flpe mice (Stock No. 009086) to remove the neo cassette. Resulting miR-206^flox floxed offspring, lacking the neo cassette, were backcrossed to C57BL/6J mice for at least 30 generations by the donating laboratory. Upon arrival, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes.
