ChgaCreER-2A-tdTomato knock-in/knock-out mice express an inducible cre/ERT2 fusion gene and TdTomato under the control of the endogenous Chga promoter. The insertion of the cre/ERT2-2A-tdTomato sequence replaces 56 base pairs downstream of the ATG start codon of the Chga gene. Chga encodes a marker of mature enteroendocrine cells (EECs), specialized endoderm derived epithelial cells located throughout the gastrointestinal tract. EECs act as the guts primary chemosensors, monitoring content and releasing hormones to regulate digestion, appetite, and gut-brain communication. When these ChgaCreER-2A-tdTomato mice are bred with mice containing loxP-flanked sequences, tamoxifen-inducible Cre-mediated recombination will result in deletion of floxed sequences in mature enteroendocrine cells (EECs) located throughout the gastrointestinal tract. Mice that are heterozygous for this knock-in/knock-out allele are viable and fertile. The donating investigator did not make a homozygous colony. No Cre activity is evident in these mice prior to tamoxifen administration. TdTomato expression is detectable by immunofluorescence staining, direct microscopy, and FACS analysis.
