Description

Dusp2^stopFl mice contain Cre-inducible Dusp2 and EGFP sequences with a ubiquitously expressed synthetic CAG promoter inserted into the Gt(ROSA)26Sor locus. After removal of the loxP-flanked STOP cassette, DUSP2 and EGFP are expressed by Cre-expressing cells. These mice may be useful when studying lymphoid cell proliferation and differentiation.

Detailed description

Dusp2^stopFl mice contain Cre-inducible Dusp2 and EGFP sequences with a ubiquitously expressed synthetic CAG promoter inserted into the Gt(ROSA)26Sor locus. A floxed STOP cassette, containing a neo cassette, prevents expression of the Dusp2 gene and EGFP in the absence of Cre recombinase. After removal of the loxP-flanked STOP cassette, DUSP2 protein and EGFP are produced in Cre-expressing cells. The donating investigator reports no EGFP fluorescence prior to Cre recombination.

Dusp2 encodes a nuclear dual-specific threonine/tyrosine phosphatase restricted to immune cells. Its expression is induced by antigenic and mitogenic stimulation in various immune cells, such as T cells, B cells, mast cells, and other leukocytes. DUSP2 has been implicated in the promotion of immune cell proliferation and malignant transformation.

Dusp2^stopFl mice were bred to Cγ1-cre mice (Stock No. 010611) to overexpress Dusp2 in B lymphocytes. After cre expression, Dusp2 overexpression promotes B-cell proliferation and malignant transformation by regulating cell cycle progression via enhancing the activation of cyclin dependent kinase 1 (CDK1). After cre expression, EGFP is detectable by immunohistochemistry and FACS analysis.

Development

The Dusp2^stopFl targeting vector contains a floxed stop cassette, containing a neomycin resistance (neo) cassette, upstream of the coding sequence for the murine dual specificity phosphatase 2 (Dusp2) cDNA, an IRES sequence, and an EGFP sequence driven by a CAG promoter. This targeting vector also included 5' and 3' homology arms for site-directed recombination into the Gt(ROSA)26Sor locus. The construct was electroporated into C57BL/6J-derived embryonic stem (ES) cells and correctly targeted ES cells were injected into blastocysts. The resulting chimeric mice were bred to C57BL/6J mice (Stock No. 000664) for at least 5 generations by the donating laboratory. Upon arrival at The Jackson Laboratory, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes.
