Tac1 encodes the preprotachykinin A precursor protein which is further processed into four neuropeptides, Substance P (SP), neurokinin A (NKA), neuropeptide K (NPK) and neuropeptide &gamma;. The primary peptides generated, SP and NKA, are involved with pain transmission, inflammation, neurogenic responses and smooth muscle contraction. Tac1-IRES-Flp knock-in (KI) mice have a FLP recombinase (flp) gene inserted into the 3' UTR of teh Tac1 gene, while retaining endogenous Tac1 expression. This insertion is designed to have the endogenous promoter/enhancer elements direct flp expression to tachykinin 1-positive cells. Heterozygous and homozygous Tac1-IRES-Flp mice are viable and fertile with no overt phenotype. When Tac1-IRES-Flp KI mice are bred with mice containing frt-flanked sequences, Flp-mediated recombination will result in deletion of the frt-flanked sequences in Tac1-expressing cells in the offspring. The donating investigator reports Flp recombinase expression in TAC1-positive cells of the dorsal vagal complex (DVC) in the medulla by measuring EGFP expression after injection of an AAV-fDIO-EGFP. These Tac1-IRES-Flp KI mice may be useful when studying nociceptive pathways, neurobehavioral processes, and neurogenic inflammation.
