IFN-brite knock-in mice have two copies of the mGreenLantern (mGL) reporter sequence inserted just upstream of the stop codon of the ISG15 ubiquitin-like modifier (Isg15) gene. mGreenLantern encodes a bright monomeric mutant GFP with a maturation half-time of 13.5 minutes. Each of the mGreenLantern sequences were preceded with a 2A self-cleaving peptide which allows for separate expression of mGreenLantern and ISG15 in these mice. Isg15 encodes a ubiquitin-like protein that is a highly abundant interferon stimulated gene (ISG) detected in many cell types, including immune, stromal, and neuronal cells. Using in vivo and in vitro cytokine delivery or viral infection, mGreenLantern fluorescence is evident in IFN-brite mice even at low levels of IFN signaling across cell types including immune cells, stromal cells, and neurons throughout the body, including lung epithelium and brain. Fluorescence is detectable via direct fluorescence???, immunofluorescence, and FACS. The reporter is highly sensitive to Type I signaling, with lesser affinity for Type II signaling. Homozygous mice are viable and fertile.
