Description

The Trpv1-floxed allele has loxP sites flanking exon 2 of the mouse Trpv1 gene, which encodes a capsaicin- and heat-activated ion channel, enabling Cre recombinase-dependent Trpv1 knock-out. These mice may be useful in studies of peripheral nociceptors and various inflammatory and neuropathic pain conditions.

Detailed description

Trpv1, a capsaicin- and heat-activated ion channel, is expressed by peripheral nociceptors and has been implicated in various inflammatory and neuropathic pain conditions. The TRPV1-floxed mice have a conditional, Cre recombinase-dependent knock-out of the mouse Trpv1 gene. Prior to the introduction of Cre recombinase, mice heterozygous or homozygous for the Trpv1^flox allele are viable and fertile with normal breeding and have no gross physical or behavioral abnormalities. When Trpv1^f/f are bred to mice that express Cre recombinase, the resulting offspring will have exon 2 deleted, creating a null allele in the cells/tissues as determined by the Cre promoter.

Of note, the Jackson Laboratory has the Trpv1 KO allele-carrying mice donated by the same investigator (Stock No. 003770).

Development

Two loxP sites were inserted by the CRISPR/Cas9 system across the second exon of the Trpv1 gene (Reference transcript: ENSMUST00000102526.9), where the ATG codon is located. Two crRNAs (5'-UCA AGG UGU CCU GAU UAA CGG UUU UAG AGC UAU GCU-3' and 5'-CCA CAC UCU UGC GAU CUU GCG UUU UAG AGC UAU GCU-3') were separately annealed to tracrRNA. The annealed oligos and Cas9 nuclease forming ribonucleoprotein (RNP) complexes and a repair template were microinjected into 129 strain mouse embryos. Mice carrying the targeted loxP insertions were identified by PCR and confirmed by sequencing. One confirmed founder was crossed to C57BL/6J inbred mice for at least five generations, and was maintained on a C57BL/6J genetic background by the donating laboratory (see SNP note below). Upon arrival, sperm were cryopreserved. Cryorecovery/live colony development will involve C57BL/6J (Stock No. 000664).



In 2026, a 48 SNP (single nucleotide polymorphism) panel analysis, with 43 markers covering all 19 chromosomes and the X chromosome, as well as 5 markers that distinguish between the C57BL/6J and C57BL/6N substrains, was performed on the males sent to The Jackson Laboratory Repository. Six of the 43 markers throughout the genome were segregating with 129. Also, three markers throughout the genome were segregating with an unknown source. These data suggest the mice sent to The Jackson Laboratory Repository were on a mixed genetic background.
