Description

Ifng^L floxed mice possess loxP sites flanking exons 2-3 of the Ifng gene. These mice may be useful when studying the role of interferon-gamma (IFN-γ) in innate and adaptive immunity.

Detailed description

Ifng (interferon gamma) encodes a cytokine secreted by cells of the immune system and is involved in cellular response to viral and microbial infections. Aberrant expression of IFN-γ is associated with autoimmune disease. Ifng^L mice have loxP sites flanking exons 2-3 of the Ifng gene.

Heterozygous and homozygous mice are viable and fertile with no overt phenotype. When bred to mice that express tissue-specific Cre recombinase, resulting offspring will have exons 2-3 deleted, resulting in complete loss of IFN-γ in the cre-expressing tissues. This floxed mouse strain may be useful in studies of immune response, inflammation and autoimmunity.

Development

The Ifng^L allele was generated using CRISPR/Cas9 endonuclease-mediated genome editing. Guide RNAs (5'-GCTGTTTCCCTGCGTAGTTT-3'and 5'-TAGAGGCTAACCAGAGCCGA-3') were used to introduce loxP sites, along with BamHI sites, flanking exons 2-3 of the interferon gamma (Ifng) gene on chromosome 10. The sgRNA, ssDNA, and Cas9 nuclease were introduced into the cytoplasm of zygotes derived from C57BL/6 mice with well-recognized pronuclei. Correctly targeted pups were identified by PCR and sequencing. The resulting Ifng^L floxed mice were maintained on a C57BL/6J background for more than 20 generations by the donating laboratory. To establish a live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J (Stock No. 000664) oocytes.
