Description

These Tg(Pdgfrb-Cre)35Vli transgenic mice express cre recombinase in Pdgfrb-expressing cells. When these mice are bred with mice containing a loxP-flanked sequence of interest, Cre-mediated recombination will result in deletion of the floxed sequences in Pdgfrb-expressing cells of the offspring.

Of note, PDGFRβ-P2A-CreER^T2 mice expressing Cre-ER^T2 from the endogenous Pdgfrb promoter are available as Stock No. 030201.

Detailed description

The Tg(Pdgfrb-cre)35Vli mice express a nuclear targeted Cre recombinase driven by the mouse platelet derived growth factor receptor, beta polypeptide (Pdgfrb) promoter. When these transgenic mice are bred with mice containing loxP-flanked sequences, Cre-recombinase activity is detected in Pdgfrb-expressing cells of the offspring. The donating laboratory bred these Tg(Pdgfrb-cre)35Vli mice to a LacZ-reporter mouse line (B6;129S4-Gt(ROSA)26Sor^tm1Sor/J) and confirmed that β-galactosidase activity was consistent with endogenous Pdgfrb expression in resulting progeny.

Hemizygous mice are viable and fertile. Homozygous mice are not viable. This strain may be useful for understanding the role of PDGFRB in in blood vessel development, and the migration and recruitment of pericytes and smooth muscle cells.

Development

The Tg(Pdgfrb-Cre)35Vli transgenic construct was generated by cloning and ligating a 4.8kb fragment, including the first exon, of mouse platelet derived growth factor receptor, beta polypeptide (Pdgfrb) promoter sequence to a nuclear targeted Cre recombinase sequence. The construct was microinjected into fertilized oocytes from FVB/N x B6;129S4-Gt(ROSA)26Sor^tm1Sor/J matings. Six founder lines were generated, with founder line 35 selected for further expansion. Tg(Pdgfrb-Cre)35Vli were backcrossed to C57BL/6J for at least 11 generations by the donating laboratory (see SNP note below). Upon arrival, sperm was cryopreserved. To establish a live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes.

Of note, the transgene integration site and copy number have not been confirmed to date.

In 2025, a 48 SNP (single nucleotide polymorphism) panel analysis, with 43 markers covering all 19 chromosomes and the X chromosome, as well as 5 markers that distinguish between the C57BL/6J and C57BL/6N substrains, was performed on the males sent to The Jackson Laboratory Repository. Two of the 43 markers throughout the genome were segregating with 129 or FVB, or C57BL/6 substrain.
